At the time of the Beit Hamikdash (the Temple), everyone had to set aside one piece of the dough from the bread that was made and gave it to the Cohanim (the high priests), who, being at G-d’s permanent service, could thus feed themselves without having the slightest effort to provide.

Since the Beit Hamikdash has been destroyed, Challa is separated by all those who knead the dough (including Cohanim) with one difference - because the Cohanim no longer perform their priestly activities, this piece of dough is now destroyed.
